Mechanism

How VIP Works

VIP binds VPAC1 and VPAC2 G-protein-coupled receptors, activating adenylate cyclase and increasing intracellular cAMP. This cascade suppresses NF-κB activation and reduces production of TNF-α, IL-6, IL-12, and IL-18 while upregulating anti-inflammatory IL-10. VIP promotes regulatory T-cell (Treg) differentiation, inhibits Th1/Th17 polarisation, and protects neurons via BDNF and PACAP-related pathways. In the lung, it acts as a potent bronchodilator and vasodilator via smooth muscle relaxation.
