Mechanism

How Melittin Works

Melittin inserts into phospholipid bilayers, forming toroidal pores and causing rapid membrane depolarisation and lysis. In cancer research, it activates apoptotic pathways (caspase-3/9 activation, cytochrome c release), inhibits NF-κB and MAPK signalling, and demonstrates anti-angiogenic effects by reducing VEGF expression. Its antimicrobial activity operates through the same membrane-disruption mechanism, active against gram-positive and gram-negative bacteria, enveloped viruses (including HIV and HCV), and drug-resistant pathogens. Targeted nanoparticle conjugates are under active investigation for tumour-selective delivery.
