VIP (Vasoactive Intestinal Peptide) is one of the most potent anti-inflammatory signalling molecules in the body, reducing TNF-alpha, IL-6, IL-12, and nitric oxide production in activated macrophages and dendritic cells. It also promotes regulatory T-cell differentiation, shifting the immune balance toward tolerance and resolution.
MOTS-c activates AMPK (the master metabolic regulator) promoting fat oxidation, glucose uptake, and mitochondrial biogenesis. It also translocates to the nucleus under metabolic stress to coordinate antioxidant gene programs. Together with VIP's immune modulation, it creates conditions for both energy restoration and immune balance.
The nervous and immune systems are tightly coupled through shared peptide signals. VIP is produced by neurons and has receptors on immune cells; MOTS-c is produced by mitochondria and influences both metabolic and immune function. Side effects reflect both component peptides. VIP may produce mild transient hypotension (vasodilation) or facial flushing at higher doses due to its vasoactive properties: administering slowly and remaining recumbent briefly after injection minimises this. MOTS-c is generally very well-tolerated. Mild fatigue during metabolic adaptation may occur in the first days. The combination is not recommended for individuals with pre-existing hypotension. No organ toxicity or hormonal effects have been documented for either component.
