SLU-PP-332 binds to and activates ERRα and ERRγ, transcription factors that control PGC-1alpha-dependent gene programs involved in mitochondrial biogenesis, fatty-acid oxidation, oxidative-phosphorylation enzyme expression, and cardiac and skeletal-muscle oxidative capacity. Activation of these pathways resembles the molecular signature of aerobic training: increased mitochondrial number and efficiency, improved fat oxidation, enhanced cardiac output, and reduced metabolic markers of aging. Preclinical studies have reported improved aerobic capacity, reduced adiposity, and improved metabolic parameters without changes in activity level.

ERRα and ERRγ are master transcriptional regulators of mitochondrial biogenesis via the PGC-1alpha pathway. SLU-PP-332 directly activates these receptors, upregulating the transcription of genes that govern new mitochondria formation: increasing the density and capacity of the cellular energy infrastructure in skeletal muscle, cardiac tissue, and metabolically active cells.
SLU-PP-332 activates molecular gene programs normally associated with sustained aerobic training, including PGC-1alpha targets, oxidative-phosphorylation enzymes, and fatty-acid transport and oxidation genes.
