Humanin

Mitochondria-Derived Cytoprotective Peptide

Humanin is a mitochondria-derived survival peptide that protects cells from dying prematurely, shields neurons from Alzheimer's-related damage, and reduces arterial plaque formation. Research shows offspring of centenarians carry two to three times more Humanin than their peers, suggesting it is part of the biology of exceptional longevity.

Mechanism

How Humanin Works

Humanin exerts cytoprotective effects through multiple mechanisms. It binds to and inhibits the pro-apoptotic protein BAX, preventing mitochondrial outer membrane permeabilisation and downstream caspase activation. It also activates the JAK2/STAT3 survival pathway, promotes AMPK-mediated metabolic adaptation, and reduces oxidative stress by enhancing antioxidant gene expression. In the vasculature, Humanin reduces endothelial inflammation and foam cell formation, providing atherosclerotic protection. In the nervous system, it protects neurons from amyloid-beta toxicity and oxidative insult.
