Like Selank, AC-Selank-NH2 modulates GABA-A receptor sensitivity, elevates BDNF, normalises serotonin and dopamine turnover, and reduces cortisol-mediated stress signalling. The acetyl modification protects the N-terminus from aminopeptidase degradation, extending plasma half-life and CNS residence time. The amide modification at the C-terminus confers resistance to carboxypeptidase activity. Together, these modifications allow a lower effective dose to achieve greater receptor engagement, producing more profound anxiolytic, anti-stress, and neuroplasticity effects compared to unmodified Selank, with the same freedom from sedation, tolerance, and dependence.

The acetylation of the N-terminus and amidation of the C-terminus in AC-Selank-NH2 significantly increase the molecule's resistance to enzymatic degradation and its affinity for target receptors. The result is a more potent interaction with GABA-A receptor modulatory sites and the neuropeptide receptors that mediate Selank's anxiolytic effects.
Standard Selank's relatively short plasma half-life limits its duration of action. The AC-NH2 modifications block aminopeptidase and carboxypeptidase cleavage, extending both plasma half-life and CNS residence time: enabling a single dose to maintain anxiolytic and neuroplasticity effects for significantly longer than the parent molecule.
AC-Selank-NH2 elevates brain-derived neurotrophic factor (BDNF) expression more robustly and for longer than standard Selank, supporting neuroplasticity, memory consolidation, and antidepressant-like effects, without any GABAergic sedation. Users maintain full alertness and cognitive function throughout the anxiolytic effect.
Unlike benzodiazepines, which globally enhance GABA-A activity and cause sedation, tolerance, and dependence, AC-Selank-NH2 modulates GABA-A receptor sensitivity in a more nuanced, receptor-subtype-selective manner. This produces calm and reduced anxiety without the sedative or amnestic properties that characterise conventional anxiolytic drugs.
Chronic stress dysregulates the hypothalamic-pituitary-adrenal axis, resulting in cortisol hypersecretion and disrupted circadian cortisol patterns. AC-Selank-NH2 normalises both ACTH secretion and the downstream cortisol response to acute stressors: reducing the physiological substrate of anxiety and restoring resilience to stress exposure.
Selank's anti-inflammatory CNS effects (mediated via IL-6, TNF-alpha, and NFkB pathway modulation) are preserved and extended in the AC-NH2 form. This neuroprotective dimension makes AC-Selank-NH2 relevant not only for anxiety management but for any protocol where reducing neuroinflammatory tone is a goal.

AC-Selank-NH2 is very well tolerated, sharing the favourable safety profile of standard Selank with no meaningful adverse effect additions from the terminal modifications. No sedation, no cognitive impairment, no tolerance formation, and no withdrawal effects have been documented. Mild injection site reactions are the most commonly noted observation. No organ toxicity, cardiovascular effects, or hormonal interference have been attributed to Selank in any research. Its non-benzodiazepine mechanism makes it categorically safer for anxiolytic use than pharmacological alternatives.
